Dysregulation of protein s palmitoylation can disrupt cellular pathways and contribute to the development of various diseases, particularly cancers. aberrant s palmitoylation has been extensively studied and proven to be involved in tumor initiation and growth, metastasis, and apoptosis. Additionally, we discuss the methods to detect s palmitoylation, the limitations of available dhhc targeting inhibitors, and ongoing research efforts to address these obstacles. Therefore, targeting palmitoylation is a novel therapeutic approach for treating human immunologic diseases. in this review, we discuss the role that palmitoylation plays in both immunity and immunologic diseases as well as the significant potential of targeting palmitoylation in disease treatment. Mutation of the cys teine of the dhhc motif can block the auto s palmitoylation and transfer activity of zdhhcs, suggesting that the cysteine of the dhhc motif is necessary for s palmitoylation [39, 40]. however, this catalytic motif is not strictly limited to elements composed of dhhc amino acids.
