Microbiologists use a variety of different media to grow microorganisms, and we will be using a wide variety of these media over the course of the semester, including some media that selects for or differentiates between the growth of specific types of bacteria. A common method for the isolation of a pure culture from a mixture is by "streaking" plates. the inoculum is streaked over the agar surface to isolate colonies on at least a portion of the plate. The streak plate technique is used to isolate the organisms (mostly bacteria) from a mixed population into a pure culture. the inoculum is streaked over the agar surface to “thin out” the bacteria. Starting the streak on the side of the plate farthest from your dominant hand, pass the loop on the surface of the agar in a zig zag pattern filling the surface of the plate. see figure to the right. replace the lid, and immediately flame the loop. place the plate upside down for incubation. Understand the basic tenets of the streak plate technique; and correctly use the streak plate technique to isolate discrete bacterial colonies.
